Externally Funded Doctoral Student (m/f/d) - Epigenetic Regulation of Pancreatic Cancer Cell Plasticity (Johnsen group) Full-time Tätigkeitsfeld: Wissenschaft und Forschung Beschäftigungsumfang: Vollzeit Company Description The research group of Prof. Steven Johnsen is part of the Robert Bosch Center for Tumor Diseases (RBCT). The RBCT is a newly founded non-profit translational cancer research institute at the Bosch Health Campus in Stuttgart, in direct proximity to the Robert Bosch Hospital (RBK). As a part of the National Center for Tumor Diseases (NCT) Southwest and the Comprehensive Cancer Center Tübingen-Stuttgart, the RBCT is dedicated to innovative cancer research and the development of new treatment strategies. The Johnsen group has a particular and long-standing expertise in the elucidation of complex gene regulatory networks that control pancreatic cancer pathogenesis. In particular, we utilize state-of-the-art techniques such as various genome-/transcriptome-wide sequencing approaches, high-throughput CRISPR/Cas9-mediated screening, multiplex immunofluorescence staining of tissues, single cell RNA-sequencing, as well as genetic engineering and other work on tumor cell lines, primary patient samples and translational tumor models. Our main research goal is to identify gene regulatory mechanisms that can be exploited as novel therapeutic targets to improve treatment response and pancreatic cancer patient survival. Job Description Utilize modern sequencing-based molecular biology approaches (RNA-seq, ChIP-seq, HiChIP, Hi-C, PRO-seq, etc.) to test hypothesis and examine epigenetic and gene regulatory mechanisms controlling pancreatic tumor cell identity and therapy response. Bioinformatic analysis and interpretation of data followed by hypothesis testing and generation. Perform multiplex immunofluorescence staining to confirm gene regulatory mechanisms in primary tumor tissue using multispectral imaging with an Akoya PhenoImager. Perform tissue-based preparation of histological samples (FFPE tumor material) for single cell RNA-sequencing using the 10X Genomics technology. Perform advanced cell culture experiments to examine the effects of drug treatment in various translational model systems. Participation in internal or external research collaborations.
